PNC-27

Overview

PNC-27 is a 32–amino acid chimeric peptide composed of two functional regions: residues 12–26 of the p53 transactivation domain, which contains the HDM-2 binding motif, fused to a membrane-residency penetratin-like sequence at the C-terminus.

Research Background

PNC-27 has been studied in oncology research focused on selective targeting of cancer cells. Published studies report its activity on a range of solid-tumor lines and certain leukemia models that express HDM-2 in their plasma membrane.

Storage & Handling

Store lyophilized peptide at -20°C for long-term stability. Once reconstituted with bacteriostatic water, store at 2–8°C and use within recommended timeframes. Handle under sterile laboratory conditions and avoid repeated freeze-thaw cycles.

PNC-27 binds membrane-associated HDM-2 (the human homolog of MDM-2), which is expressed on the plasma membrane of many transformed cells but is largely absent from non-transformed cell membranes. The p53-derived sequence adopts an HDM-2-binding conformation, and the C-terminal membrane-residency peptide drives transmembrane pore formation. The combined binding and pore formation lead to selective membranolysis of cells expressing membrane HDM-2, sparing normal cells in published research models.

PNC-27 is studied in oncology research, membrane-biology research, and HDM-2/p53 axis pharmacology. It is a reference compound in p53-derived chimeric peptide research.
